Reference (0142) To a mixture of 1.00 g of 2-amino-3-methylbenzoic acid, 8 mL of acetic acid, 4 mL of 50percent hydrobromic acid and 16 mL of water were added 0.46 g of sodium nitrite and 3 mL of water under ice cooling. After stirring the mixture for 10 minutes under ice cooling, disappearance of 2-amino-3-methylbenzoic acid was confirmed by silica gel thin-layer chromatography (developing solvent: hexane/ethyl acetate=1/1, Rf value of 2-amino-3-methylbenzoic acid: 0.67), then 1.42 g of copper bromide and 5 mL of water were added, and the mixture was heated at 50° C. for 3 hours. Water was added to the mixture, and the resulting mixture was filtered to obtain 1.0 g of 2-bromo-3-methylbenzoic acid. (0143)
